Sherman Klump, un énorme professeur de chimie dans un collège californien, se désole de sa surcharge pondérale, d'autant plus qu'on vient de lui adjoindre une jeune et sympathique collègue qu'il aimerait bien séduire. Il travaille depuis longtemps à une formule capable de faire fondre les graisses. Devant l'urgence, il n'hésite pas à tester sa découverte sur lui-même. Il se transforme immédiatement en un irrésistible bellâtre, mais il s'aperçoit que son invention n'a pas que des effets bénéfiques.
